What is PE-22-28?

PE-22-28 is a shortened version of spadin, a peptide from the sortilin precursor protein. It blocks the potassium channel TREK-1, which has been linked to depression, and is therefore promoted as a fast-acting antidepressant and nootropic.

How well studied is PE-22-28?

Data are limited to cell and mouse experiments in which shortened spadin analogues inhibited TREK-1 more strongly and were active in depression models. No human studies are known.

What side effects and risks does PE-22-28 have?

Effects and side effects in humans are unknown. TREK-1 is also found in the heart and other tissues; the consequences of blocking it outside the brain have not been studied. Self-treating depression with untested substances can delay effective therapy.

Is PE-22-28 approved in Germany?

In short: no. Not approved anywhere, no known clinical development. In Germany an unapproved substance (AMG); prohibited by WADA under S0 as a non-approved substance. Grey-market products are untested.
